Arbeitsgebiet
Dr. Oliver Pötz entwickelt multiplexe Sandwich Immunoassays und kombinierte Immunoaffinitäts-Massenspektrometrie-Assays für die Proteomforschung. Beide Ansätze haben technologische Limitierungen und können nicht gleichzeitig einen hohen Grad an Multiplex bei gleichzeitiger hohen Sensitivität und Probendurchsatz erreichen. Beide Methoden sind komplementär und werden je nach biologischer Fragestellung eingesetzt. Multiplexe Sandwich Immunoassays können für wenige dutzend Analyte aufgebaut werden und stellen sensitive, durchsatzfähige Assaysysteme dar. Massenspektrometrie basierte Methoden sind per se multiplex und es können eine Vielzahl von Proteinen aus einer Probe identifiziert werden, allerdings ist eine aufwändige Probenfraktionierung notwendig, um entsprechende Sensitivität erreichen zu können. Mittels einer speziellen Immunoaffinitätsanreicherungsmethode – TXP Proteomics – können pro Antikörper mehrere Signaturpeptide angereichert werden und anschließend per Massenspektrometrie identifiziert werden.
Dr. Pötz entwickelt neue Methoden für die Analyse von Signalwegen in Tumorgeweben und Zellkulturassays.
